Sephadex is a trademark for dextran cross-linked gel used for gel filtration. Launched by Pharmacia in 1959, after development work by Jerker Porath and Per Flodin. This name comes from se parasi Pha rmacia dex tran. Usually produced in bead shape and most often used for gel filtration columns. By varying the degree of crosslinking, gel fractionation properties can be altered.

This highly specialized gel filtration and media chromatography consists of synthetic macroscopic beads derived from dextran polysaccharides. The organic chain is crosslinked to provide a three-dimensional network having a functional ion group attached by the ether linkage to the polysaccharide chain glucose unit.

Available forms include anion and cation exchangers, as well as gel filtration resins, with varying degrees of porosity; bead sizes fall within the discrete range between 20 and 300 Âμm.

Sephadex is also used for ion-exange chromatography.

Apps

Sephadex is used to separate molecules of low molecular weight and height. Sephadex is a faster alternative to de-salting, requiring a low dilution factor (as small as 1.4: 1), with high activity recovery. Sephadex is also used for exchange of buffers and the removal of small molecules during the preparation of large biomolecules, such as ampholytes, detergents, radioactive or fluorescent labels, and phenols (during DNA purification).
